      为了明确解淀粉芽孢杆菌B9601-Y2(下称Y2)的促生长机制,通过摇瓶培养,检测其溶磷解钾固氮活性,通过盆栽试验,探索Y2对玉米生长量与溶磷解钾量的影响。结果表明Y2及其绿色荧光标记菌GFP-Y2(下称GFP-Y2)均具有良好的溶磷解钾固氮活性,Y2、GFP-Y2与克雷伯氏菌Yn固氮量分别为2.9mg/L、2.7mg/L、3.3mg/L;Y2、Y2-GFP与洋葱伯克霍尔德氏菌1-2在溶磷培养基培养4天的悬浮液中,溶磷量达732μg/mL、711μg/mL和758μg/mL;Y2、GFP-Y2与胶质类芽孢杆菌G2在钾长石培养基中,解钾量分别达13.5?g/mL、12.42?g/mL、17.17?g/mL。Y2、GFP-Y2和G2发酵液均能显著增加玉米生长量,分别增加玉米型高28.29%、31.01%、24.42%,根长27.21%、30.15%、25.74%,叶宽18.56%、17.37%和13.17%,鲜重50%-80%,干重70%-95%。Y2发酵液处理后土壤中速效氮、速效磷和速效钾含量分别增加了20.42%、111.01%和17.24%;Y2-GFP处理速效氮磷钾分别增加了16.33%、83.33%和19.98%,而空白对照及LB处理的速效氮磷钾含量在播种后均较播种前低。此外Y2、Y2-GFP和G2发酵液的施入可使植株氮含量较空白对照分别增加,较LB培养基处理增加37.07%、32.35%、19.33%,磷含量也显著高于空白对照和LB处理,分别增长90%-120%和70%-100%不等,这说明功能菌活化了土壤难溶性磷、钾,促进了植株对氮、磷及钾素营养的吸收利用,从而促进了玉米的生长。本研究为Y2微生物菌肥的开发提供了实验依据。

      To understand the plant-growth promotion mechanisms and the phosphorate-, postassium-dissolving, and nitrogen-fixing(PPN) activities of of Bacillus amylolyquefaciens B9601-Y2(Y2),the activities were tested in shaking culture and the growth promoting and PPN activities were studied by pot experiment of maize. The results showed that Y2 and its GFP-labelled strain, GFP-Y2, had good PPN activities. Y2, GFP-Y2 and Klebsiella sp. Yn could fix 2.9mg/L,2.7mg/L and 3.3mg/L of nitrogen in non-nitrogen medium, Y2, GFP-Y2 and Burkholderia cepaciea 1-2 could release 732μg/mL,711μg/mL and 758μg/mL of available phosphorus(AP) in phosphate dissolving medium, Y2, GFP-Y2 and Paenibacillus mucilaginosus G2 could release 13.5?g/mL,12.42?g/mL and 17.17?g/mL of available potassium(AK) in fledspar dissolving medium by 4-days culturing. Y2, GFP-Y2 and G2 fermentations could increase significantly biomass of maize campared with the negative control (CK), i.e. 28.29%,31.01%and 24.42% of type height,27.21%,30.15%,25.74% of root length,18.56%,17.37% and 13.17% of leaf width, 50%-80% of fresh weight, 70%-95% of dry weight. Compared with the soil before the treatments, the soil increased 0.42% available nitrogen, 111.01% AP and 17.24% AK after Y2 application and 16.33%,83.33% and 19.98%, respectively, after Y2-GFP application. The AN, AP and AK, however, reduced in the soilstreated withCKand LB medium. Drenching Y2, GFP-Y2 and G2 fermentation liquids could increase 45.46%,40.44% and 26.63% total nitrogen of maize plant more than CK, 37.07%,32.35% and 19.33% more than LB medium application, 90%-120% and 70%-100% of total phosphorus more than CK and LB medium application, respectively. This indicated that the beneficial microbes activated combined phosphorus and potassium, and promote maize plants to absorb N, P and K, and grow. This study may provide the Y2 strain with the basis to formulate biofertilizer.
