      摘要:快速碱化因子(RALF)是一种能够参与植物发育调控的植物肽激素,在真菌中也广泛存在并且影响着真菌对寄主的侵染状况。本文通过外源添加酸碱来明确pH值对玉米大斑菌生长的影响,并使用实时荧光定量PCR技术来分析玉米大斑病菌中StRALF基因在外源添加酸与碱时表达量的变化。运用预测网站对克隆得到的StRALF基因编码区蛋白序列的基本性质进行分析。结果表明,玉米大斑菌在碱性环境下生长状态较好;碱性条件下,StRALF基因比酸性条件下基因表达量高。在侵染寄主过程中,96h及以后表达量达到最大;经预测分析,初步确定其为分泌蛋白或膜蛋白。这说明在弱碱性条件下,StRALF基因表达量较高并参与侵染过程以及侵染后的生长阶段。本文为RALF基因在玉米大斑病菌中的作用及作用机制的深入研究提供参考。

      Abstract: Rapid alkalization factor (RALF) is a plant peptide hormone that can partici pate in the regulation of plant development. It also exists widely in fungi and affects the infection of fungi to the host. In this paper, the effects of pH value on the growth of Setosphaeria turcica were determined by adding acid and alkali. And the changes in the expression of StRALF gene in the Setosphaeria turc ica were analyzed by real -time fluorescence quantitative PCR technology. The basic properties of StRALF gene coding region protein sequence were analyzed by prediction website. The results showed that the growth state of Setosphaeria turcica was better in alkaline environme nt; In alkaline condition, the expression of StRALF gene was higher than that in acid conditi on. In the process of host infection, the expression of 96 h and later reached the max imum, and it was preliminarily determined as secretory protein or membrane protein by prediction analysis. Above description, under the weak alkaline condition, the expression level of StRALF gene was high and involved in the infection process and the growth stage after infection. This study can provide a reference for the further research on the action and action mechanism of RALF in the Setosphaeria turcica.
