摘要:【目的】明确玉米黑粉菌菌瘤内内生细菌种类及其对玉米黑粉菌冬孢子萌发的影响。【方法】利用高通量测序分析明确玉米黑粉菌菌瘤内菌粉培养液中内生细菌种类,结合16SrDNA分析,分离并鉴定在培养液中占比最高的菌株。以溶菌酶及Amp处理玉米黑粉菌冬孢子为对比,显微镜下观察菌瘤内内生细菌对玉米黑粉菌冬孢子萌发的影响。【结果】高通量测序分析表明在4份样品的菌粉培养液中,肠杆菌属细菌的占比在95%以上,当肠杆菌大量增殖时对黑粉菌的冬孢子萌发产生影响,可以引起黑粉菌的冬孢子发生质壁分离,释放原生质体,部分原生质体再生成菌体;也可以使冬孢子壁直接被降解,原生质体萎缩。【结论】本研究发现玉米黑粉菌菌瘤内的内生细菌以肠杆菌为主,该内生细菌通过分泌类似溶菌酶的物质对黑粉菌的冬孢子产生影响,其对玉米黑粉菌初侵染具有促进作用,为研究该类内生细菌与黑粉菌的作用机制及其在玉米黑粉菌侵染过程中作用打下基础。

Abstract:[Objective] To investigate the endophytic bacteria species in corn smut and their effects on the germination of brandspore of Ustilago maydis.[Methods]High-throughput sequencing analysis was used to identify the dominant endophytic bacteria in the culture medium of corn smut gall. We isolated the strain with the highest proportion in the culture medium and identified it with 16SrDNA analysis. Comparing the effects of lysozyme, Ampicillin and endophytic bacteria on brandspores of Ustilago maydis, we observed and analyzed their differences through the microscope. [Results]High-throughput sequencing analysis showed that in the culture medium of four samples, the dominant bacterias are unclassified-Enterobacteriaceae,belonging to Enterobacteriaceae, accounted for more than 95%.While the Enterobacter proliferate in large numbers, the endophytic bacteria can affect the germination of brandspore of Ustilago maydis by causing the brandspore of Ustilago maydis to be separated from the plasma wall and release protoplasts. Meanwhile, some of the protoplasts regenerated into mycelium. Also, there were some brandspore to be degraded directly and some protoplasts to become atrophy. [Conclusion] The dominant bacterias are unclassified-Enterobacteriaceae in corn smut. During the growth and reproduction process, Enterobacter in the corn smut may secrete substances similar to lysozyme to affect the brandspore of Ustilago maydis. Therefore, this study is a foundation for the study of the interaction mechanism between and Ustilago maydis, and the role of the Enterobacteriaceae in the infection process of Ustilago maydis.
