引起玉米叶部病害的玉米大斑病菌(Setosphaeria turcica) 是一种异宗配合型真菌,其基因组内含有两种关键的交配型基因 StMAT1-1 和 StMAT1-2, 其存在和功能直接影响玉米大斑病菌的有性生殖能力。大量研究显示交配型基因对于细胞凋亡、DNA 修复和细胞增殖等生命过程至关重要,但其作用机制尚未得到明确解析。利用本试验室先前建立的玉米大斑病菌 cDNA文库,构建诱饵载体pGBKT7-StMAT1-1和pGBKT7-StMAT1-2,利用酵母双杂交筛玉米大斑病菌cDNA文库,进一步通过酵母双杂交一对一试验验证互作蛋白,共计得到StMAT1-1互作蛋白6个和StMAT1-2互作蛋白4个。这些候选互作蛋白质的功能部分与细胞分裂增殖相关,部分可以为有性生殖提供能量,通过与MAT蛋白互作在有性生殖过程中发挥作用。本研究为进一步研究 StMAT1-1和StMAT1-2作用机制奠定基础,也为深入研究有性生殖网络提供理论基础。

Setosphaeria turcica, the causal agent of corn leaf blight, is an allopatric mating-type fungus whose genome contains two key mating-type genes, StMAT1-1 and StMAT1-2. Their presence and function directly influence the fungus's capacity for sexual reproduction. Extensive studies indicate that mating-type genes are crucial for vital processes such as apoptosis, DNA repair, and cell proliferation, yet their underlying mechanisms remain poorly understood. Using the S. turcica cDNA library previously established in our laboratory, we constructed bait vectors pGBKT7- StMAT1-1and pGBKT7-StMAT1-2. We screened S. turcica cDNA library using yeast two-hybrid technology, sequenced the obtained positive clones, and validated the genes through BLAST searches on the NCBI website. Gene functions were then annotated. Further validation of interacting proteins was conducted through one-to-one yeast two-hybrid assays, identifying six StMAT1-1 interacting proteins and four StMAT1-2 interacting proteins. The functions of these candidate interacting proteins are partially associated with cell division and proliferation, and partially capable of providing energy for sexual reproduction. They exert their roles in the process of sexual reproduction through interacting with MAT proteins. This study lays the foundation for further investigation into the mechanisms of StMAT1-1 and StMAT1-2 and provides a theoretical basis for in-depth research on sexual reproduction networks.
