LysM家族基因包括含LysM受体样蛋白(LYP)和含LysM受体样激酶(LYK),参与识别几丁质、脂多糖和肽聚糖以触发植物免疫反应。为揭示LysM基因家族在玉米生长发育及抗病中的作用,利用生物信息学方法对玉米LysM基因家族成员进行了鉴定,并分析了其蛋白的理化性质、亚细胞定位、系统发育关系及基因启动子区的顺式作用元件。利用RNA-Seq及qRT-PCR技术分析了LysM家族成员在玉米不同组织和大斑病菌侵染时的表达模式。结果显示,在玉米中共存在11个LysM家族基因ZmLysM1~ ZmLysM11,这11个LysM基因家族成员不均匀地分布在7条染色体上;通过亚细胞定位预测发现,11个LysM基因家族成员编码的蛋白在质膜、叶绿体、胞外均有分布;系统发育分析发现,玉米LysM家族成员与单子叶植物水稻和小麦具有较近的亲缘关系;顺式作用元件分析发现,该家族成员启动子序列存在多个激素响应、逆境胁迫响应和光响应元件;通过RNA-Seq数据分析发现,有10个玉米LysM家族成员至少在一个玉米组织中表达,其中ZmLysM1、ZmLysM4和ZmLysM6呈组成性表达模式,ZmLysM10只在根组织中表达,ZmLysM5、ZmLysM7和ZmLysM8在部分组织特异表达;qRT-PCR分析显示,ZmLysM2、ZmLysM6、ZmLysM7在玉米大斑病菌侵染后差异表达,推测其可能在响应大斑病菌侵染过程中发挥作用。本研究明确了玉米LysM家族基因的结构特征及表达模式,可为后续揭示该家族基因的功能及作用机制提供参考。

LysM gene family includes the LysM receptor-like protein (LYP) and LysM receptor-like kinase (LYK) proteins. These proteins are involved in the recognition of chitin, lipopolysaccharide and peptidoglycan to trigger the plant immune system. To elucidate the role of LysM gene family in maize growth, development and disease resistance, the members of LysM gene family were identified in maize using bioinformatics methods, and the physicochemical properties, subcellular localization and phylogenetic relationships of the their proteins were analyzed. The cis-acting elements in the promoter region of the LysM genes were analyzed. The expression patterns of the LysM family members were analyzed in different tissues and in response to Setosphearia turcica infection using RNA-Seq and qRT-PCR, respectively. The results showed that there were 11 LysM genes in maize, named as ZmLysM1~ ZmLysM11. The 11 LysM genes were unevenly distributed on seven maize chromosomes. Subcellular localization prediction revealed that 11 LysM proteins were distributed in the plasma membrane, chloroplast, and extracellular. Phylogenetic analysis revealed that the maize LysM family members were closely related to rice and wheat which were belong to monocotyledons. Cis acting element analysis identified several hormone responsive, stress responsive and light responsive elements in the promoter sequences of LysM genes. Analysis of RNA-Seq data showed that 10 maize LysMs were expressed in at least one maize tissue, among which, ZmLysM1, ZmLysM4 and ZmLysM6 showed a constitutive expression pattern, ZmLysM10 was expressed only in root tissue, and ZmLysM5, ZmLysM7 and ZmLysM8 were partially tissue-specific expressed. qRT-PCR analysis showed that ZmLysM2, ZmLysM6, and ZmLysM7 were differentially expressed after inoculation with S. turcica, suggesting a role in the response to S. turcica infection. In the present study, we identify the structural features and expression patterns of maize LysM family genes, and these result will provide insight into subsequent functional and mechanistic studies of members of the LysM gene family.
